摘要 | Compared to commonly used in vitro blood-retinal-barrier (BRB) models that utilize transwells to establish barriers, a microfluidic-based BRB model provides more in vivo-like microenvironments to prompt the formation of tight junctions (TJs) for permeability screening of drug candidates. Here, we developed a microfluidic-based BRB model system for drug permeability study. An in vitro BRB model is built by co-culturing retina pigment epithelial (RPE) cells and choroid endothelial cells on a porous polycarbonate membrane, where the measured trans-epithelial-endothelial resistance (TEER) of the barrier increased up to 45 kΩ•mm. This device is believed to bring an applicable strategy for in vitro ophthalmic drug permeability tests. |
