摘要 | Zoledronic acid is an antitumor drug. STAT3 signaling pathway is positively correlated with non-small cell lung cancer (NSCLC). However, whether zoledronic acid affects the proliferation and apoptosis of NSCLC through STAT3 signaling pathway is unclear. The control group (NC group), zoledronic acid group (ZOLEDA group), and zoledronic acid + epidermal growth factor EGF group (ZOLEDA + EGF group) were set up followed by analysis of STAT3 mRNA level by qRT-PCR; p-STAT3, STAT3, Cyclin D1, Bcl-2, and Tubulin protein expression by Western blot, cell proliferation by clone formation and MTT assay, and cell apoptosis by Annexin V-FITC/PI staining. Compared with NC group, ZOLEDA group showed significantly reduced p-STAT3/STAT3 ratio and expression of Cyclin D1, and Bcl-2 protein (P < 0.01), which were significantly increased in ZOLEDA + EGF group (P < 0.05); clone formation and MTT results showed that compared with NC group, cell proliferation in ZOLEDA group was significantly reduced (P < 0.01), and increased in ZOLEDA + EGF group (P < 0.05). In addition, ZOLEDA group presented significantly promoted cell apoptosis (P < 0.01), which was decreased in ZOLEDA + EGF group (P < 0.05). Zoledronic acid (ZOLEDA) can inhibit STAT3 signaling pathway and cell proliferation and promote cell apoptosis. STAT3 signaling pathway activator EGF can prevent the effect of zoledronic acid. |
