摘要 | After a myocardial infarction (MI), the border zone (BZ) is characterized by an oxygen (O2) gradient located at the boundary between the injured, hypoxic cardiac tissue and adjacent normoxic, viable tissue. In this study, underlying microfluidic channels were used to generate and expose engineered cardiac tissues to stable O2 gradients as a “Border-Zone-on-a-Chip” model. Functional and transcriptomic analysis revealed that calcium transients, calcium wave propagation velocity (CPV), contractile stress, and gene expression were uniquely regulated by an O2 gradient when compared to engineered tissues exposed to global, homogeneous O2 levels. |
