Statement of Purpose: Glycolysis and oxidative phosphorylation are the two principal pathways in cell metabolism. Measurements of acidic metabolites and consumption of oxygen in culture medium caused by cell metabolism and respiration are significant for evaluating glycolysis and oxidative phosphorylation. Abnormal glycolysis and oxidative phosphorylation are related to various diseases, such as diabetes, Alzheimer’s disease and cancers. Therefore, developing dual pH and oxygen nanosensors is crucial for cell metabolism understanding, high-throughput analysis and diseases diagnosis. However, dual pH and oxygen optical sensors developed in previous studies are less sensitive and show lower quantum efficiency. Herein, we prepared amphiphilic graft copolymer-based dual luminescent pH and oxygen nanosensors with high sensitivity and quantum efficiency in extracellular cell metabolism monitoring.
